EDCI coupling of 2-bromopropionic acid with ammonia solution to form MD 20-1 that can then be coupled with 1-(4-fluorophenyl)-2,5-dimethyl-1H-pyrrole-3-carboxylic acid.

HOBt (0.086 g, 0.64 mmol) and EDCI (0.11 g, 6.0 mmol, 1.2 equiv.) were added to a solution of 2-bromopropionic acid (0.45 mL,) in DCM (5 mL) after 20 minutes of stirring, after which 28% Ammonia(aq) (0.28 mL, 7.5 mmol, 1.5 equiv.) was added and the reaction stirred at room temperature.
